Effects of Dicyclohexylcarbodimede on the Microsomal ATPase Activity of the Rat Brain

Abstract
Both Mg" and Na+-K+ ATPase activities in microsomal fraction obtained from rat brain cortex were measured in vitro under the variety of conditions. The effect of dicyclohexylcarbodiimide(DCCD) on the activities of both ATPases was investigated and the results were summarized as follows.
1. Both Mg" and Na+-K+ ATPase activities were depressed markedly by DCCD.
2. The inhibition of Na+-K+ ATPase activity of microsomal fraction of the brain cortex by DCCD increased in lower pH and in the presence of hydroxylamine, while the inhibition of Mg* ATPase activity by DCCD was not related with pH change.
3. The inhibition of Na+-K+ ATPase activity by DCCD was not prevented by an addition of cysteine in the medium.
4. The inhibition of Na+-K+ ATPase activity by DCCD was protected markedly by the presence of Na-*- but not K+.
These results indicate that Na+-K+ activated ATPase is a multienzyme system, and remarkable corre¡©lation between activation of ATPase and carboxyl groups could be observed.
